In the second episode, an infected Sam met one of the late Death's subordinates and received a disturbing warning. He and his brother will not be able to get resurrected the next time they die.
Fortunately, clever as he was, the younger Winchester was eventually able to determine that holy fire can cleanse him from the mysterious virus.
Nevertheless, they still need to find Darkness (Amara) who apparently eats human souls. In order to even remotely understand the ancient evil, the hunters would have to ask the angels and demons.
That plan doesn't seem to be sound however with Crowley (Mark Sheppard) wanting Amara for himself and Castiel (Misha Collins) still weighed down by Rowena's attack dog spell.

It was her who helped take the Mark of Cain off of Dean but according to the "Supernatural" season 11 spoilers that executive producer Jeremy Carver shared, her newfound powers will pose as a problem for the brothers.
"We'll be seeing her in the early parts of the season deciding what she wants to do with this newfound power," he told TV Line.
Castiel and Crowley may know that Darkness is dangerous and powerful but nobody knows exactly what kind of destruction she'll bring to Earth.
Jensen Ackles previously told Entertainment Weekly that "whatever the reach of that character, what that character can do, we're not quite sure of," adding that they "don't know the scope of her powers."
The second episode already showed her capable of starting a "Biblically unclean" infection and sucking souls out of people, Sam and Dean will need to speed up their effort to put a stop to Darkness.
